Therefore, one could consider a model in which λ plasmid replication carried out by the heritable replication complex is unidirectional (either leftward or rightward depending on the parental DNA strand which inherited the replication complex), and that carried out by a newly assembled complex is bidirectional, thus giving a mixture of unidirectionally and bidirectionally replicating molecules in a population of plasmids maintained in the host cells growing under standard laboratory conditions.

To determine which of these two alternative possibilities is true, we investigated directionality of λ plasmid replication in E.coli relA + hosts and relA mutants growing under normal conditions and in the amino acid-starved relA mutant, i.e. under conditions in which only the replication carried out by the heritable complex could occur.
